ABSTRACT

Objective: to investigate the relation between genotoxicity and interleukin-6 in rats exposed to cigarette smoke


Study Design: an experimental study


Place and Duration of Study: army Medical College, Rawalpindi and Armed Forces Institute of Pathology, Rawalpindi, in 2016


Methodology: seventy healthy Sprague Dawley rats were placed in smoke chambers at animal house of National Institute of Health, Islamabad. Cigarette smoke was given to them for 3 months. Genotoxicity was assessed by Cytokinesis Block Micronucleus [CBMN] assay. Enzyme linked immunosorbent assay [ELISA] kit was used to determine Interleukin-6 in study samples. Pearson correlation was used to find the correlation between genotoxicity and IL-6


Results: the mean IL-6 and micronuclei frequency was 49.48 +/- 19.69 ng/L and 6.77 +/- 0.73, respectively. Weak positive association was found between micronuclei frequency and IL-6 in smoke exposed rats [r=0.266, N=70, p=0.026]


Conclusion: genotoxicity and inflammation are associated in smokers. The present study concluded that smoke exposure elicited a proinflammatory profile, which might have promoted DNA damage in smokers

ABSTRACT

Objective: To investigate the relation between serum amyloid A-low density lipoprotein [SAA-LDL] and genotoxicity in smokers


Study Design: An experimental study


Place and Duration of Study: Army Medical College, Rawalpindi and National Institute of Health [NIH], Islamabad, from June 2014 to February 2015


Methodology: Seventy healthy Sprague Dawley rats were purchased from NIH and exposed to cigarette smoke in smoke chamber for three months. Blood samples were drawn from each rat at the end of the study period. SAA-LDL was determined by enzyme-linked immunosorbent assay [ELISA]. Genotoxicity was assessed by cytokinesis block micronucleus [CBMN] assay. Pearson correlation was used to find correlation between SAA-LDL and genotoxicity


Results: Strong positive correlation was found between SAA-LDL and micronuclei frequency in smoke-exposed rats [r=0.799, N=70, p <0.01]


Conclusion: Statistically significant strong positive correlation between SAA-LDL and genotoxicity in smoke-exposed rats shows that changes in one is associated with changes in other and vice versa

ABSTRACT

Objective: To investigate the effect of cigarette smoking on interleukin-6 in Sprague Dawley rats. Study Design: Randomized controlled study. Place and Duration of Study: Study was conducted at Army Medical College Rawalpindi, from Jan to Apr 2015


Material and Methods: Seventy healthy male Sprague Dawley rats were randomly divided into two groups at National Institute of Health [NIH]. Group-I rats were not exposed while group-II rats were exposed to cigarette smoke for 3 months. Interleukin-6 [IL-6] was determined using commercially available enzyme linked immunosorbent assay [ELISA] kit. Data was analyzed using t-test


Results: Higher levels of IL-6 were observed in exposed than in non-exposed. The mean of IL-6 levels of control and smoker groups was 40.84 +/- 9.09 and 49.48 +/- 19.69 respectively. The difference was statistically significant [p<0.05]


Conclusion: Positive association was found between cigarette smoking and IL-6. This shows that IL-6 may be used as a biochemical marker to assess inflammatory damage in smoke related diseases

ABSTRACT

Background: Tribulus terrestris is a creeping herb that has been used in the past for treating impotence. The extract obtained from Tribulus mainly contains the active component protodioscin. Administration of the extract improves libido and spermatogenesis. Protodioscin increases the levels of luteinizing hormone [LH], testosterone, dehydroepiandrosterone [DHEA] and dihydrotestosterone [DHT]. The objective of present study was to determine the effect of Tribulus terrestris on serum luteinizing hormone in male Sprague Dawley rats


Methods: The study was carried on 60 adult male Sprague Dawley rats aged 90-120 days and with average body weight of 200+/-50 grams. Rats were divided into two groups with 30 rats per group. Group A received 0.5 ml distilled water once daily orally. Group B was administered orally Tribulus aqueous extract 6 mg/Kg once daily. All the doses were given to rats for 8 weeks. After 8 weeks, serum luteinizing hormone was measured in both groups. The mean values of LH were compared using SPSS. Alpha value was kept at 0.05


Results: The mean LH level was 1.26+/-0.54 for control group while it was 1.75+/-0.65 for the experimental group. The difference between control group and experimental group was statistically significant [p<0.001]


Conclusion: Tribulus terrestris increases the levels of LH when administered in appropriate doses
